Lithium Chloride Precipitation Solution is formulated 7.5 M LiCl, 50 mM EDTA colorless solutions for RNA isolation and in vitro transcription. A final concentration of 2.5M LiCl is effective in precipitation of RNA larger than 300 nucleotides without the addition of ethanol, this method of precipitation selectively precipitates RNA and does not efficiently precipitate DNA, protein or carbohydrate.
The basic principle involves the interaction between ribose sugar of RNA and positive cations (lithium) to allow the folding and precipitation of RNA only, leaving out the DNA and protein in the aqueous solution. The final precipitated RNA can be collected after removing out the aqueous (unprecipitated) sample.
In spite of the above-mentioned advantages, this technique might not be suitable for isolating the lower concentration of RNAs and different types of RNA other than mRNAs, such as tRNA and may not work as well for RNAs under ~ 100 nucleotides.Quote request
